(i) Explant : Plant part that is excised from its original location and used in initiating a culture. It may be shoot tip, root tip, shoot bud, anther, embryo, ovule etc. Normally undifferentiated and meristematic cells of plants are used as explant.

(ii) Surface sterilization : The process by which explants are treated with specific antimicrobial chemical like H2O2, sodium hypochloride, Bromine water, C2H5OH etc.

(iii) Sterlization : The vessel, media and instruments are also suitably treated with steam, dry heat or alcohol or subjected to alteration to make them free from microbes.
