The full form of LPS in Medical is Lipopolysaccharide. It is the common element that is found on the outer membrane of Gram-negative bacteria. It acts as an immunostimulatory agent triggering the release of pro-inflammatory cytokines. LPS comprises three elements that help to maintain the structural integrity of the bacteria. It leads to inflammation, cytokinin shock, and septic shock for the host cell if not treated on time.

Components of LPS

LPS is a complex protein that is found in the outer membrane of Gram-negative bacteria. It consists of three major components: lipid A, core oligosaccharide, and O antigen. LPS plays a crucial role in the structural integrity of bacterial cell membranes. Here are the major details related to its components:
  • Lipid A is the innermost portion of LPS and is embedded in the bacterial membrane. This is composed of a phosphorylated glucosamine backbone along with fatty acid chains. Lipid A is highly hydrophobic and anchors the LPS molecule within the bacterial outer membrane. Lipid A offers endotoxic properties which eventually trigger the immune response leading to triggering fever, inflammation, and other symptoms
  • The core oligosaccharide is a shorter carbohydrate chain that extends from the lipid A region. It connects lipid A to the O antigen and can vary in length and composition between different bacterial species. This helps to maintain the structural integrity of the LPS molecule
  • The O antigen is the outermost portion of LPS and consists of a repeating chain of sugar molecules. This portion of LPS varies extensively between bacterial strains and is used for bacterial serotyping and classification. The O antigen is responsible for the antigenic diversity found in different Gram-negative bacterial species

Features of LPS

  • LPS is known for its potent endotoxin activity allows it to offer a strong immune response in the host. When LPS is released from bacterial cells inside it leads to the production of cytokines, inflammation, and fever
  • The O antigen (O polysaccharide) component of LPS varies widely among different bacterial strains and species. This variability leads to differences in antigenic characteristics. This is extremely helpful as scientists use these variations for bacterial identification and classification
  • It plays a critical role in maintaining the structural integrity and stability of the bacterial cell envelope. The lipid A region anchors LPS in the lipid bilayer of the outer membrane, while the O antigen extends beyond the bacterial surface
  • LPS contributes to the barrier function of the bacterial outer membrane. It helps to regulate the passage of molecules, ions, and nutrients across the membrane

Function of LPS

LPS are integral components of Gram-negative bacterial outer membranes. It mainly comprises three elements namely, lipid A, core oligosaccharide, and O antigen. The major role of LPA is to regulate cell integrity and helps to regulate the permeability of the membrane. Also, LPS whenever comes in contact with a toll-like receptor induces inflammation and aids pathogen recognition. 

Disadvantages of LPS

LPS is found in the cell membrane of bacteria and thus offers a range of disadvantages to the human body. The following are the major risks associated with the LPS.
  • LPS is a potent activator of the host immune response. When activated in excess, it can lead to inflammation, cytokine storm, septic shock, and multiple organ failure
  • In the absence of proper medication, LPS can lead to septic shock that can result in low blood pressure, organ dysfunction, and a high mortality rate
  • Chronic exposure to low levels of LPS can lead to the development of chronic inflammatory disorders and can cause damage to the tissues
  • LPS activates the immune system and can potentially trigger autoimmune responses that attack the body's own tissues.
  • The lipid A component leads to the release of large amounts of lipid A into the bloodstream can lead to endotoxic shock, organ dysfunction, and a high risk of mortality
  • Creating effective vaccines targeting LPS is difficult due to the varied diversity of different bacterial strains and species
